Above all, however, the NMDA-antagonistic functional mechanisms prove to be particularly effective in Parkinson's disease. These NMDA antagonists inhibit the effects of glutamate. This messenger substance is particularly present in high concentrations in Parkinson's sufferers. Overall, budipine must therefore be assigned to the dopamine receptor agonists, the MAO inhibitors and the NMDA receptor antagonists.
Pharmacological effect
The available drugs against Parkinson's disease can ensure that the imbalance of various messenger substances in the brain can be balanced. In this way, they help to enable information to be transferred from one cell to another. The mechanism of action of Budipin is only partially known. The agent penetrates the blood-brain barrier.
On the one hand, it stimulates the dopamine receptors, mimicking the effects of dopamine. On the other hand, the active ingredient ensures that the neurotransmitter glutamate is only released in an inhibited manner. This prevents the uptake of dopamine in the nerve cells. Budipin also influences the enzyme monoamine oxidase (MAO), which means that dopamine can also have a reinforcing effect.
Medicines with Budipin favor the influence of almost all neurotransmitter systems in the brain that are impaired in Parkinson's disease. The two main effects of the remedy include a reduction in tremor and an effect that promotes movement. Clinical data on the effectiveness of the agent are only available to a limited extent. The half-life of the active ingredient is 31 hours.
A detailed examination of the heart is necessary before starting therapy. Medicines containing budipine can cause life-threatening cardiac arrhythmias in individual cases. Due to these negative effects on the heart, patients must sign a written declaration of commitment to regular ECG checks before starting therapy. An ECG must be written not only at the beginning of therapy, but also after one week and three weeks after the initial medication. An examination should then be carried out at least once a year.
Medical application & use
Budipin is particularly suitable for treating minor illnesses. The active ingredient is also suitable for a combination treatment with other Parkinson's drugs, so that it can also be used in more advanced stages. In mildly pronounced cases, therapy alone can also be sought. Therapy usually begins with a dose of 10 mg, whereby the dose can be increased up to 3x20 mg. The maximum daily dose is 30mg three times.
Parkinson's treatment with Budipin should be started gradually. By slowly increasing the dose, side effects can be kept as low as possible. The remedy should be taken in the morning or during the morning. Use after 4 p.m. should be avoided, as the late medication increases the risk of possible sleep disorders. In order to avoid life-threatening conditions such as ventricular fibrillation or cardiac arrhythmias, the contraindications must be strictly observed during treatment. Both ECG checks and an adjustment of the electrolyte balance must be carried out.
If patients experience their heart unusually quickly and irregularly (palpitations), complain of dizziness or brief loss of consciousness, the drug must be discontinued immediately. In the case of a targeted withdrawal, the agent must be slowly tapered off.
The origin of Parkinson's disease has not yet been sufficiently clarified. Targeted prevention is not possible. However, both a healthy lifestyle and adequate exercise and mental activity can likely delay the onset of the disease.

Dizziness as well as restlessness and fatigue can also occur. Other negative side effects include headaches, loss of appetite, hot flashes and visual disturbances. In some cases, both nightmares and confusion and hallucinations can occur.
Budipin may only be prescribed by doctors who stipulate in writing that they strictly adhere to the prescribed precautionary measures. In addition to regular ECG checks and with strict consideration of the contraindications, this also includes balancing the electrolytes.
Budipin may not be taken under certain circumstances. In addition to cardiac insufficiency, cardiac arrhythmias (including AV block and ventricular arrhythmias), cardiac deceleration (bradycardia) and cardiac muscle diseases, the contraindications include the neuromuscular disorder myasthenia gravis and potassium and magnesium deficiency.
